Structural Changes in HPRT Gene of V79 Cells After Irradiation With Heavy Ions—Immediate and Delayed Effects

چکیده
The radiobiological effects of accelerated ions with high charge and energy (HZE) on mammalian cells their propagation in time are still not sufficiently explained attract great deal attention. This work aims to compare the immediate delayed emphasis latter. As shown by our group, dependence mutant fraction expression after irradiation may have interesting, non-monotonic, character depending LET (linear transfer) used heavy ions. We speculate that this phenomenon occur due induced genomic instability. Another area research is study DNA structural changes these mutants at different times. Chinese hamster V79 were irradiated 11 B, 18 O, 20 Ne, gamma radiation. was ranging from 0.23 keV/?m 60 Co rays up 136 Ne unique HPRT isolated, concentration measured, exons amplified, analyzed several points, about 40 days, exposure. Over 1200 for deletions sorted into three main categories: partial deletion, PD—with deletion one eight exons; total deletions, TD—with all nine deleted; no deletions—no change structure observed. In general, number samples increasing radiation, suggesting higher deposition cell nucleus more likely cause larger changes. case increase observed ?115 followed a sharp decrease. also distribution particular various times, so-called mutational patterns. Hypothesis mechanisms behind phenomena given, possible implications further discussed.

Background: All aerobically growing organisms suffer from exposure to oxidative stress, caused by partially reduced forms of molecular oxygen, known as reactive oxygen species (ROS). These are highly reactive and capable of damaging cellular constituents such as DNA, lipids and proteins. Consequently, cells from many different organisms have evolved mechanisms to protect their components...

15 صفحه اولAnalysis of mutations in the human HPRT gene induced by accelerated heavy-ion irradiation.
Multiplex PCR analysis of HPRT(-) mutations in human embryo (HE) cells induced by 230 keV/microns carbon-ion irradiation showed no large deletion around the exon regions of the locus gene in contrast to the irradiations at different LETs. To identify these mutations, the sequence alterations in a cDNA of hprt gene were determined for 18 mutant clones in this study. Missing of exon 6 was the mos...
متن کاملon translation of politeness strategies in dialogues involving female characters in translations and retranslations of novels translated before and after the islamic revolution of iran and their effects on the image of women: a polysystem theory approach
abstract reception environment has considerable effects on accepting a translation. as the expectations of a target culture and its values and needs change throughout history, its criteria for accepting a translation or rejecting it will change accordingly (gentzler, 2001). the expectations of iran, as the reception environment in the present study, have changed after the islamic revolution. i...
